Drug discovery is under increasing pressure to deliver increasing numbers of development candidates to fill the corporate drug pipeline. This means that there is an urgent need for high quality lead compounds in order to drive the provision of candidates for pre-clinical development. This problem is compounded by the ever-increasing number of putative therapeutic targets coming out of Genomic and Proteomic research. Our computational and medicinal chemists work closely together utilizing our proprietary suite of programs in the design of novel scaffolds that interact optimally with the chosen target binding sites. These scaffolds are further elaborated with appropriate reagents inÊsilico, to produce virtual combinatorial libraries that can be readily synthesised. It is the close collaboration between Computational, Medicinal and Combinatorial Chemists that makes this process so efficient.
Our comprehensive computational chemistry and structural biology services help you get the most from your synthesis and screening programs. We believe that optimal lead discovery and optimization is the result of a continual thorough analysis and understanding of all relevant structural, chemical, and biological information. Our proprietary methods enable this critical understanding of the target, its ligands, and the target-ligand relationship. When the structure of the target is known we employ proprietary structure based design methods. Our Token™, technology is used to characterize and compare protein surfaces. Through this analysis we can design for affinity and specificity reducing the likelihood of undesirable crossreactivity and late stage clinical failure. Using SurFlex™ Dock and Refinery™ we are also able to rapidly perform and analyze virtual screening experiments to identify meaningful leads for biological screening. When the structure of the target is not known, we employ proprietary ligand based design methods to design and rapidly analyze the results of high throughput screening experiments. Initial screening compounds may be selected to be similar to known ligands, or maximally diverse when little or no ligand information is available. Screening results are rapidly analyzed using Support Vector Machine algorithms which are able to determine which features of a molecule correlate best with activity, and those which correlate with inactivity. This knowledge is then used to formulate a Structure Activity Relationship which guides compound synthesis for follow on screening.

The Bridge Between Genomics and Medicine Will Be Built on Proteomics. Traditional methods of drug development involve screening thousands of chemical compounds. Compounds are first tested in cell culture, and those that show the desired activity are tested in animal models. Finally, the few compounds that pass animal testing are taken into human clinical trials. Only about one in 10,000 chemical compounds makes it through this laborious process. In contrast, LifeSpan's gene localization technologies help pharmaceutical companies to rapidly identify the most promising drug targets. By localizing proteins and messenger RNAs within human tissue samples, LifeSpan can determine when and where individual genes are expressed - down to the subcellular level. By reducing the number of potential drug targets to genes for which expression is altered with disease, new drugs can be developed more quickly and cost-effectively. Sequencing the human genome was a milestone for science. But it was just the first step in a longer journey toward understanding disease processes. That's where LifeSpan helps.

Protein Pathways is a privately held biotechnology company, founded in 1999, that is a leader in the development of algorithms that can be applied to the functional interpretation of sequenced genomes. Our proprietary system of pathway elucidation is called the ProMaps™ discovery system. While most comparative genomics analyses are based almost exclusively on similarity searches that infer only limited aspects of biological function, the ProMaps™ discovery system can predict gene function, place newly discovered genes in existing pathways and elucidate interactions between pathways. In other words, by using the ProMaps™ discovery system it is possible not only to predict the biological classification of a gene product, but to also gain insight into interacting proteins, the nature and identification of probable substrates, and the biological context of the associated pathway. The company has established the discovery of molecular interactions as its core competency because of the firm belief that placing genes in existing pathways and understanding the network interactions between pathways will inevitably lead to improved methods that facilitate the small molecule drug discovery process.

Proteom's technology enables the intelligent selection of targets and ligands in silico before entering costly drug discovery programmes in the laboratory.
The Proteome Factory AG is a modern biotech company. Scientific research focuses on neurodegenerative disease and oncology. The proteomic key technology platform enables key proteins in emerging disease and biomarkers for the disease state to be rapidly and efficiently identified. Target proteins are then validated, initially through the use of virally and stably transfected cell lines and, at a later stage, transgenic and knock-out technologies are applied. This combination of validatory techniques enables the Proteome Factory AG to distinguish more relevant target proteins for a specific disease process, highlighting possible therapeutic targets. The main task of the Proteome Factory AG therefore is the development and validation of new therapeutic drugs with a particular focus on neurodegenerative disease and cancer.

The Spendlove Research Foundation was founded in 1989 and is funded through an endowment established by Dr. Rex S. and Reta Spendlove. Dr. Spendlove is the Founder of Hyclone Laboratories, a leading producer and distributor of cell culture media products throughout the world. From its inception up until 1997 the Spendlove Research Foundation supported outside researchers through capital investments and other forms of financial support. In March of 1997 the Spendlove Research Foundation changed its strategy to concentrating on and supporting its own research projects. This has allowed the Spendlove Research Foundation to control both the direction and the emphasis of various research and development projects. The Spendlove Research Foundation is located in beautiful Cache Valley in Northern Utah near Utah State University in Logan. The close proximity to the University allows the Foundation to hire highly trained and qualified individuals who desire to continue their education in the pursuit of advanced degrees. Our long-term research goals focus in the area of in vivo cell culture simulation. This research centers on specialty cell culture media that will eventually allow researchers to control the activities of the cell through the media and cell environment. In order to achieve that end, we are developing special analytical systems to allow us to measure the biochemical components in blood, organs, and cells. We are in the business of technology development and transfer. We develop unique and beneficial technologies in-house and out-license them for commercialization. In order to increase the probability of commercialization success, we will generally give narrow rather than broad licenses - licensing the technology to many companies, to sell to many markets.

Zyomyx has developed technology that enables proteins to be analyzed with the kind of precision and throughput that automated sequencers and DNA chips brought to the study of our genes. Not just an adaptation of DNA chip technology - the Zyomyx platform was designed from the ground up for proteins. A result of over 250 man-years of focused research and development, it is the only platform to deliver the needed combination of biological specificity, high sample throughput, sensitivity, and massive parallelism. The power of Zyomyx' technology will be utilized by pharmaceutical and biotech companies to validate the backlog of targets created by their genomics programs, to study the mechanism of action for new drug leads, and to enable real time monitoring of therapeutic response and potential side effects. It will also enable diagnostics companies to move quickly, identify new biological markers, and enable clinicians to run hundreds of tests from a single drop of blood or small tissue samples. Given the flexibility and broad applicability of the technology there will likely be numerous other applications throughout life science research and in other fields. Zyomyx will profit by supplying proteomics research tools and by collaborating with others to apply them in pharmaceutical research. It will also supply technology and components to other companies pursuing complementary applications.
